Deep phenotyping seborrheic dermatitis patients


Elucidate novel disease characteristics and biomarkers. Understand pathogenesis in respect to inflammation vs. Malassezia vs. microbiome. Unravel skin barrier function and lipids in-depth

The complex pathophysiology of immunological skin diseases requires a novel, method-richand integrated approach. Therefore, multidimensional profiling of diseases by means of systems pharmacology / dermatology is necessary. Over the recent yearCHDR-LACDR-LUMC-TNO have developed a comprehensive methodology to phenotype patients in various dimensions including molecular and cellular information from the skin tissue, the cutaneous microbiome, clinical assessments from the physician and patient-reported outcomes.

Complementary techniques are available at LACDRwith the lipidomics platform (LC-MS analysis) and biophysical characterization(FTIR). Aim of the project is the multidimensional profiling of patients and the skin for the purpose of I) getting better mechanistic understanding of disease and II) to identify and characterize biomarkers for clinical research.

At LACDR the lipidomics platform will be used to establish in this disease:

  1. Discriminationpower (lesional vs. non-lesional skin vs. healthy)
  2. Naturalcourse (lesional – placebo and non-lesional)
  3. Suitabilityas biomarker in an intervention trial
  4. Correlationto disease burden / other read outs
